Brain region specificity in reactive oxygen species production and maintenance of redox balance

چکیده
The brain produces various reactive oxygen species in enzymatic and non-enzymatic reactions as a by-product of metabolism and/or for redox signaling. Effective antioxidant system the cells maintains balance. However, neurons glia from some regions are more vulnerable to oxidative stress ischemia/reperfusion, epilepsy, neurodegenerative disorders than rest brain. Using fluorescent indicators live cell imaging confocal microscopy, we have measured rate cytosolic mitochondrial production, lipid peroxidation, glutathione levels cortex, hippocampus, midbrain, stem cerebellum acute slices rat We found that basal ROS production is at its highest cerebellum, it mainly generated by glial cells. Activation glutamate ATP led maximal rates midbrain compared Mitochondrial had only minor implication total with values cortex minimal midbrain. peroxidation was higher while GSH level similar most lowest Thus, vary across regions.

Reactive oxygen species and redox compartmentalization
Reactive oxygen species (ROS) formation and signaling are of major importance and regulate a number of processes in physiological conditions. A disruption in redox status regulation, however, has been associated with numerous pathological conditions. In recent years it has become increasingly clear that oxidative and reductive modifications are confined in a spatio-temporal manner. This makes R...
متن کاملA computational model of reactive oxygen species and redox balance in cardiac mitochondria.
Elevated levels of reactive oxygen species (ROS) play a critical role in cardiac myocyte signaling in both healthy and diseased cells. Mitochondria represent the predominant cellular source of ROS, specifically the activity of complexes I and III. The model presented here explores the modulation of electron transport chain ROS production for state 3 and state 4 respiration and the role of subst...

Objective: We have previously shown 1 ,25-dihydroxyvitamin D3 [1 ,25-(OH)2D3] to inhibit mitochondrial uncoupling protein 2 (UCP2) expression in adipocytes and that in vivo suppression of calcitriol levels with calcium-rich diets increases UCP2 expr...

Appropriate regulation of reactive oxygen species (ROS) has a significant impact on health and disease. ROS includes oxygen ions (O2̇) free radicals (superoxide [O, 2] and hydroxyl radicals), and peroxides (hydrogen peroxide [H2O2]) and are the products of normal oxygen consuming metabolic process in the body. ROS are small and highly reactive molecules with important cell signaling roles when m...

Seizure activity has been proposed to result in the generation of reactive oxygen species (ROS), which then contribute to seizure-induced neuronal damage and eventually cell death. Although the mechanisms of seizure-induced ROS generation are unclear, mitochondria and cellular calcium overload have been proposed to have a crucial role. We aim to determine the sources of seizure-induced ROS and ...
